In a similar vein to this previous thread which asks which games you beat first try on one life:

I ask, what's the toughest or longest games you ever beat the very first time you played it (ok if you peed a couple times, but no big breaks longer than a minute or two, ya know?)? Basically, in one sitting.

One game comes to mind for me--Princess Tomato in the Salad Kingdom: 15 hours.

Oh, and another: Deja Vu for NES: 5 hours

Not that it really matters how long it took, but you get the idea. The first sitting is the important part here.

i dont remember the exact time, but I remember i was home from school sick one time and I beat Shadowgate for NES in one sitting... Probably a few hrs if i remember correctly..

Super Mario Bros. 3 - played every single last level possible first sitting...took 8 hours.

The first time I played Sid Meier's Civilization, I started in the afternoon and finally became the champion of the world sometime around 8 am. If I had to guess, I'd figure on about 14 to 15 hours. On a weeknight. When I was 12 years old.

Needless to say, I didn't go to school that morning either. ;)
