When you download MAME it should have a folder called Documentation, the Mame User Guide will have all the information you need, this is a cut and paster from there:

Keys and controls

Here's a quick breakdown of the keys:
5, 6, 7, 8 Insert coin (players 1, 2, 3, 4)
1, 2, 3, 4 Start (players 1, 2, 3, 4)
Arrow keys Move
Control (left of keyboard) Button 1
Option (left of keyboard) Button 2
Spacebar Button 3
Esc Pause
Shift-Esc When paused, step one frame forward (useful for getting to exactly the right place to take a screenshot)
Command-O Return to the MacMAME window to select a different game to play
Command-Q Quit MacMAME
F2 Put the game into "service mode" (press F2 again to return to game mode)
F3 Reset the game
F4 Show graphics (use arrow keys to look through the game's graphics, F4 to return to the game)
F6 If cheats are allowed, this key toggles them on and off
F7 Load state
Shift-F7 Save state
F8 Cycle through frameskip options
F10 Toggle speed throttling (you normally want to leave this on)
F11 Toggle the frames-per-second display on and off
F12 Save a screenshot (the image file is placed in the 'Screenshots' folder)
Tab Access MAME's configuration menu, where you can adjust key settingsand dipswitches

These are MacMAME specific, and the other versions use different keys:

Player 1:

Movement - Cursor Keys
Buttons - Left Control, Left Alt, Space, Left Shift, Z, X, C, V, B (In this order, as necessary for the game)

Player 2

Movement - R, D, F, G
Buttons - A, S, Q, W (In that order... if you need more buttons, you'll have to set them yourself or use a joystick)

Exit MAME - Esc
Options Menu - Tab

When in doubt, hit Tab, and you can view/change any button assignment you want from the menu. Just be careful about changing buttons, because it lets you assign multiple buttons to different actions, or assign key combinations to an action, so if you're not careful, you might end up screwing them up. To change one of them, move to it, hit enter. When it says to hit a key, do so and then wait (If you hit other things, it'll start getting weird). You can do the same to set actions to mouse buttons, joystick buttons and so on. If you want to set two keys to an action, you can (for example) move to "Player 1 Button 1", hit enter, then hit Ctrl... then hit enter again, then hit the first joystick button. It'll then show something like "Left Ctrl OR Joystick 1 Button 1".
