Neo Mr. Do!
For me it's Mr Do!'s Castle.
The C64 version is great as well.

This question is actually tougher than I thought. A good answer could be "the one most recently played" as every game in this series is a hell of a lot of fun to play. I have to go with the original, but it's a damn close call between Castle and Do! Run Run.

I had to vote for Neo Mr. Do! due in large part to the crazy Engrish between levels. Pure comedy gold.

Do Run Run...I think it has the most strategy of the series, plus the aforementioned music is my favorite of any arcade game :) I've never played the Neo version though...

Hard choice between my two favorites: Mr. Do and Do Run Run. I pick Do Run Run. Most creative of the Mr. Do series.

Definately Mr. Do's Castle for me! That game ate SO many of my quarters back then, wait a minute... that game ate SO many of my dad's quarters back then :D

Neo Mr. Do! is "Neo" as in "Neo Geo." It's a completely different game, and IMO isn't worthy of the franchise name. It's a decent game, but the "real" Mr. Do! games are simply fantastic.
Best game? Gotta go with Do! Run Run. It's the all around best (and deepest) game of the lot, but that doesn't stop me from playing all of them equally (possibly excepting WildRide). :)

I like them ALL (and would love to see a new mr. do game), but I had to go with the original Mr. DO! as my favorite...

I like Mr. Do's Castle, but it isn't the same calibre of the original. Do Run Run makes third on my list.

Well... Neo Mr. Do! is not a COMPLETELY different game than the original, and I grudgingly agree that it is definitely an outsider to the Universal originals. Even it is not bad, but they certainly could have done a better job.
Sort of like the Twilight Zone from 2 years ago. But it does bear the name and I think it should be considered part of the series, inferior that it may be.
Adventure Mr. Do! would have been the greatest laserdisc game of all-time.
I would not count out another title featuring the clown prince of gaming, however.
I'm doing everything in my power to make sure the most unique series of all-time does not go gently into that good night!
