Besides the immortal "Cats" quote, and, quite frankly, the entire rest of the Zero Wing intro, I get a kick out of the Medi XG7 (XG5?) pilot from Colony Wars, who says in a terribly worried voice "we caaaan't let them caaaatch us." She sounds just like the chicken from Parappa the Rapper.
Speaking of Parappa, that thing is chock-full of great quotes. Quoth the Jamaican flea market frog: "In the rain and in the snow, I've got the funky funky flow."
You know, you just have to hear some of these things first hand, I guess.
But then there's House of the Dead. In fact, any [X] of the Dead.
Oh, and then there's that naughty Morrigan (text only): "My beauty can defeat brains and brawn"
In the 16-bit era and earlier, the good quotes seem to be mostly hiliarious mistranslations. Once we got 32-bit technology and voice became common, you add in delivery, too, for even more devastatingly funny results.
Yo diggity, K-dog. This topic = teh shizzle. You are the master of unlocking.

"body blow", "body blow" from the arcade punchout. Also that wacky referee in the arcade wrestling game (can't remember the name of the game but I can still hear the ref counting one......two.......threeeeeeeeeeeeee. in that really high pitch voice.) Guess these are sound effects rather than actual quotes.

That game is Exciting Hour. Emulated in MAME.
One of my favorite quotes from old games is from Bomb Squad on INTV.
If you try to input the code to diffuse the bomb and its wrong you get this shot of the skyline and then "Oh No" and then the explosion.
Something about the way they say the "Oh No" is funny as hell.

Exciting Hour was known as Mat Mania in the States.
Guardian Heroes had really good writing, which was a big surprise for a Treasure game... Sega must have touched it up quite a bit before it was released in the United States. When you're fighting in the arena as the ninja Genjiro, you'll be given the option to continue battling or quit. If you quit, Genjiro mutters, "Enough. I'm sick of wasting perfectly good throwing stars on fools."
Sega also came up with a win quote so good they used it in two fighting games, the Genesis version of Virtua Fighter 2 and Golden Axe: The Duel for the Saturn. Characters from both games would really rub in their victories with this quote: "Greetings from the land of beatings!"

I always thought Daphne's speech at the end of Dragon's Lair was funny, but not because of the words (it's the standard "Slay the dragon" bit), but because of the way she says them. You can almost hear the wind blowing between her ears...
"To slay the dragon, use the magic sword... (angelic chorus)" LOL
Maybe it's just me...
I always liked Space Ace's witty(?) banter between Kimmy and Dexter. I've died many times just because I was laughing so hard. Usually on the motorbike stage (which is hard to begin with).

"Dexter!"
"Kimmy! Jump on!"
"No thanks. I'll walk."
<Bike takes off. Kimmy jumps aboard, seated behind Ace.>
"Dexter, the Infanto Ray's about to be fired at Earth!"
"Where's the ray located?"
"In the center of the compound."
"Can you get us in there?"
"Yes."
"LUGNUTS! Does Borf know I'm here?"
"Yes, and he's geared and ready for battle!"
"What say we shoot Borf with his own ray?"
"Why yes!"
"What say we have dinner tonight?"
"Get serious."
"I am."
"Look out, Borf ol' buddy! 'cause here we come!"
"Dexter!"
"Call me Ace, huh?"
So, how'd I do from memory?

Yep that sounds right. I'll admit it doesn't sound very funny when typed out, but when you have little pink fuzz balls and jumping around it becomes much funnier.

My favorite videogame quote isn't from a game.
It happened at E3 several years ago, when the show was in Atlanta. Hasbro Interactive had just purchased Atari and they were making a big deal about it and Centipede which was going to be the first game they were releasing. Apparently Jerry Jessop had seen one of the Tramiels (I think it was Leonard) at the Hasbro booth looking around. Jerry commented something on the order of: "I saw (Leonard) Tramiel at the Hasbro booth looking at the remants of the company that he helped destroy" It was priceless!

Ninja Gaiden and Resident Evil games are good for dramatics. As for funny lines, I recall these ones....they are not always 100% exact but I try to get close.
"Are you a bad enough dude to rescue the President?" Bad Dudes. Well, most any quote from those street fighting games, Final Fight series for example, are fun to read/hear.
"You lost even though you tried to prove your manliness. Or did I just stun you with my beauty?" Erel, to Gally in story mode of Evil Zone. Actually, a lot of the characters quotes from Evil Zone are among my favorites, especially those from Erel.
As for Bionic Commando, which someone mentioned, a couple I always kind of liked were:
"Now we have you." something like that, when the enemy caught you listening in on their transmissions right before they would storm the room.
"Get out of my way, you geek/nerd." when you would bump into soldiers in some of the neutral zones, sometimes that pissed me off enough to start shooting ;)
