It may sound obvious but check that it's all there. Then check how the stuff plays. Do any of the buttons stick? Do they work? Are they too loose? Does it work? Sounds obvious and maybe even insulting but going over that should be important enough to be reminded as I doubt your getting it for a few $$.
The spring in the foot pedal and the strum bar on the guitar are the two most common faults.
They have redesigned both a bit and newer versions are supposed to be less faulty, especially the drum set not sensing hits well enough.
How much are you buying this for used? Honestly the peace of mind that buying a new one costs might be worth it, especially since you can send any faulty instrument back to EA for replacement.
This game is well worth retail, best game i've bought in years and I would buy it again. 4 player is just too much fun. <3 Rockband.
I will see if the seller has a receipt.
Will Harmonix exchange the parts or instruments if faulty without a receipt?

they've always replaced my stuff no questions asked. I've gone through 2 kick pedals now finally bought a pedal metal kit for 20 bucks to reinforce my third and it looks cool to (get em off ebay or amazon) I'd buy new also.
I also recommend drumshh pads to quiet your drumming. Also looks cool. Other than that make sure its all there and not broken.
My friend just got a used Rock Band set two weeks ago. He is sending the drum pads, kick pedal, and game disc to EA this week to be replaced, all for free. He doesn't have a receipt or anything and they are doing it, no questions asked.
Yeah there are risks in buying a used one, because the hardware definitely has it's problems, but with EA's very loose return policy I'd say it's worth the gamble to buy a used one if you are saving a bunch of money.
EA knows the components are crap and doesn't even balk when it comes to replacing them.

No proof of purchase required, no further troubleshooting....nothing. Not after the first guitar replacement, or the second.
Just watch your credit card statement to make sure they don't wrongfully charge you (finally got that fiasco fixed).

Thanks everyone for the help. I was able to pick up a mint Rockband. The seller used it only once. A few of the parts were still sealed. I havent had any problems yet, only problem is trying to force myself out of my living room.
