YuYu Hakusho Sunset Fighters (TecToy release of the Treasure game), this game was localized for South America. But since the Brasil is part of the America Continent the game should play just fine. For example, in the Majesco cardboard boxes of games it is written that the game will run on Brasil. Even if most South American releases do not have anything written most of them are compatible with North American cartridges, the SEGA CD and SEGA Saturn games were the same SEGA of America discs ...
Gangster Town if someone use a SEGA Power Base Converter and two SEGA Light Phaser guns to play this game.
NBA JAM T.E. is a nice multi player game.
Street Racer is also nice to play. But this game is not a cooperative game.
Gain Ground for the Genesis is a fun two player co-op game.
Even with just two players Micro Machines 2 on the Genesis (okay, Mega Drive, as it was only released in the UK) is utterly compulsive. Especially as you can actually plug extra joypads into the cart itself to turn it into a 3-4 player game.
Sunset Riders on SNES. The Genesis version isn't as good, but still fun to play. I also would like to second any of the Micro Machine titles. Very fun multiplayer games. Zombies Ate My Neighbors is also a great choice.

Yeah, most people have stated the obvious titles for both the SNES and the Genny. Here are a couple others:
SNES
Tetris/Dr. Mario - this is by far the best version of 2-player Tetris on any console (until Tetris Party on Wii, anyway). The classic Gameboy music is suped up and the graphics are bright and vivid!
Power Instinct - Everyone gave a shout out to Street Fighter 2 and Mortal Kombat, but this quirky tournament fighter HAS to be mentioned! Ever play a game where you fight a grandma that shoots her dentures at you? 'Nuff said!
Saturday Night Slam Masters - this is on the Genny too, but I prefer the SNES version. Technically a FOUR-player game, but great as a 2-player wrestling game.
GENESIS
Altered Beast - HELLO! This one came right out of the box! "Wise fwom your gwave!"
Forgotten Worlds - Obviously this one was forgotten by quite a few! Awesome 2-player arcade port!
I didn't play alot of 2 player games for the genesis but there was a couple.
Punisher was very fun with 2 players.
X-Men was also another game I liked with 2 players, Not sure if X-Men 2 Clones Wars is 2 players (Never played it sorry) but if it is I assume its just as good if not better then the first (I heard its better so take it for what its worth)

Yes, some of the early arcade ports to home consoles were great to play !!! Even better their still have the two players cooperative mode ...
Alien Storm is another game from SEGA that resemble the old Golden Axe formula .This time with shooting sections! To bad that those games are only on the SEGA Genesis.
One game that is on the SNES is The King of Dragons. This game is nice for two players.
Those 16-bit machines, much like other home microcomputers of that time were very nice for arcade ports ...
