The colors for the "40-column mode" are set in vdp registers... not in the video memory like the "screen mode 2". In the vdp registers, you can set the background color and the foreground color, that's all.
So, my answer is : No, there is only two colors in "40-column mode".

Well, then it would be the 40-column mode, one color for the text (screen and text identical to "Chateau Du Dragon").
But the title screen would be a problem. For the Commodore-64, it would be 200X320. On a ColecoVision, it would be 192X256. While the difference in vertical resolution isn't much to worry about, that 25% difference in horizontal resolution makes a straightforward translation out of the question.
you're right, the resolution and also the color limitation are not the same between these two system.
A monochrome picture could be a solution and use the resolution of the colecovision to do your title screen.
